Mechanism of Action (MOA)

Urea acts as a humectant and keratolytic agent. At low concentrations, it increases water content of the stratum corneum by attracting moisture. At higher concentrations, it breaks down keratin bonds in the skin, promoting exfoliation and softening of thickened skin.

Pharmacokinetics
  • Minimal systemic absorption when applied to intact skin.
  • Localized effect in epidermis; metabolized locally.
  • Any absorbed urea is excreted renally.
  • Onset: Hydrating effects within hours; keratolytic effects develop over days to weeks.
Pregnancy Category & Lactation
  • No controlled studies in humans.
  • Considered low risk due to minimal absorption.
  • Use during pregnancy and lactation only if clearly needed.
Therapeutic Class
  • Dermatologic agent
  • Keratolytic and humectant
Contraindications
