Approved Indications

Adults (Anti-Aging Use)

• Formulation: Creams, lotions, or sunscreens containing 5%–25% zinc oxide.
• Application: Apply a thin, even layer to clean, dry facial or exposed skin twice daily, preferably morning and night.
• For sun protection, apply 15–30 minutes before sun exposure and reapply every 2 hours if exposed to sunlight.

Geriatric Patients

• Use as in adults; no dose adjustment required.

Pediatric Patients

• Anti-aging use not applicable; zinc oxide use limited to diaper rash or skin protection.

Hepatic/Renal Impairment

• No dose adjustment required; minimal systemic absorption.

Special Instructions

• Apply only to intact skin.
• Do not occlude unless directed.
• Wash hands after application.

Zinc oxide acts as a physical barrier on the skin, reflecting and scattering ultraviolet radiation to protect against photoaging. It also provides a mild anti-inflammatory and antimicrobial effect, promotes epidermal healing, and enhances barrier function. These properties help prevent collagen degradation, reduce oxidative stress, and maintain hydration, contributing to a reduction in fine lines and aged skin appearance.

• Absorption: Negligible systemic absorption through intact skin.
• Distribution: Localized to stratum corneum; not systemically distributed.
• Metabolism: Not metabolized via hepatic pathways when applied topically.
• Excretion: Not systemically excreted in appreciable amounts.
• Accumulation: Not known to accumulate in tissue with normal use.

• Pregnancy: Not assigned a formal FDA category; systemic absorption is negligible. Considered safe when used topically on limited areas.
• Lactation: Unknown whether it is excreted in breast milk; use with caution. Avoid application to the breast or nipple area to prevent infant exposure.
• Recommendation: Use only as needed and avoid large-area or prolonged use during pregnancy and lactation.

• Inhalation of powder leading to respiratory irritation (rare in modern formulations).

• No significant drug-drug or drug-food interactions with topical use.
• Systemic interactions are not expected due to minimal absorption.
• Concurrent use with other topical agents may affect penetration; stagger applications if needed.
• Does not involve CYP enzyme pathways.

• No major changes in regulatory guidelines specifically for anti-aging use.
• Zinc oxide continues to be recommended in physical sunscreens for broad-spectrum UVA/UVB protection.
• Ongoing support in dermatologic guidelines for barrier repair and prevention of photoaging through routine use of zinc oxide–containing products.
